摘要
建立了HPLC法测定鸡腿菇水解液中单糖、低聚糖及多糖含量的分析方法。选用AminexHPX.87H柱,示差折光检测器,柱温50qC,5mmol/L的硫酸为流动相,流速0.4mL/min,进样量20止,外标法定量。结果表明,单糖、低聚糖及多糖均得到较好分离,其峰面积与质量浓度线性关系良好,相关系数r^2值为0.9912—0.9997,回收率为98%~106%。采用HPLC-示差法与气相色谱法测定多糖、低聚糖及单糖相比,样品无需衍生处理,过滤后可直接进样检测。该法可同时测定原料水解液中可能存在的主要单糖、低聚糖及多糖。
A high performance liquid chromatography (HPLC) method was established for determination of monosaccharides, oligosaccharide and polysaccharide in the liquors of hydrolysis on Coprinus comatus. The sample was measured by Aminex HPX-87H column(300 mm ×7.8 mm) with differential refractive index detector at 50 ℃. The mobile phase was 5 mmol/L H2SO4 with flow rate of 0. 4 mL/min and the injection volume was 20 μL. The result indicated that mixture of monosaccharides, oligosaccharide and polysaccharide was successfully separated and a good relationship was obtained between peak area and concentration of monosaccharides, oligosaccharide and polysaceharide. The correlation coefficients of those matters ranged from 0. 991 2 to 0. 999 7 and the corresponding average recoveries were in the rang of 98% - 106% . Compared with gas chromatographic method, the method could directly detect monosaccharides, oligosaccharide and polysaceharide samples after filtration without derivative processing. The novel HPLC method for the simultaneous quantification of mono- saccharides, oligosaccharide and polysaccharide is simple, rapid and accurate.
