宜宾地区蒲公英药材HPLC特征图谱定性研究 被引量:2

摘要 目的研究宜宾地区蒲公英药材HPLC特征图谱实验方法,建立蒲公英药材的HPLC特征图谱并运用HPLC-MS/MS技术对主要色谱峰进行定性分析。方法采用RP-HPLC,梯度洗脱测定10批样品。色谱条件为:Waters symmetry shieldTMC18柱(4.6mm×250mm,5μm);流动相为A为乙腈、B为0.1%磷酸,流速为1.0mL·min-1;检测波长为210nm。结果10批不同乡镇样品的特征图谱中共检出17个相对位置稳定的共有峰;通过HPLC-MS/MS技术对其中6个峰初步鉴定为:单咖啡酰酒石酸、绿原酸、咖啡酸、木犀草素葡萄糖苷、菊苣酸、木犀草素。各样品特征图谱与对照特征图谱的相似度均大于0.82。结论该方法准确、可靠,可为蒲公英药材质量控制提供科学依据。 OBJECTIVE To study the HPLC specific chromatograms testing method of Herba Taraxaci from different areas of Yibin to develope. The specific chromatogram mode and to identify the primary chromatogram peaks by HPLC-MS/MS. METHODS HPLC was utilizaed for quality assessment of 10 batches of samples. RP-HPLC analysis was performed on a Waters symmetry shieldTM C 18s column (4.6 mm × 250 ram, 5 μm)with the mixture of aeetonitrile (A) and 0.1% phosphonic acid solution(B) as mobile phase in gradient mode. The flow rate was 1.0 mL.min-1. The detective UV wavelength was 210 nm. RESULTS 17 common peaks from thc specific chromatograms of 10 batches of Herba Taraxaci were determined, 6 of which were identified by HPLC-MS/MS . The 6 peaks were as follows: caftaric acid, chlorogenic acid, caffieic acid, luteolin-O-glucoside, chicoric acid and luteolin .The similarities of the specific chromatograms of 10 batches of samples were all above 0.82. CONCLUSION This method is accurate and reliability. It could be used for quality control of Herba Taraxaci.
