二苯乙烯类化合物对肺癌细胞株生长的抑制作用 被引量:11

GROWTH INHIBITORY EFFECT OF STILBENOIDS ON LUNG CANCER CELL LINES

摘要 目的研究二苯乙烯类化合物(含3种结构类似的成分:(1)(+)-α-viniferin,(2)miyabenolC,(3)kobophenolA,一种蛋白激酶C抑制剂)对人肺癌细胞株A549,95-D,SPC-A-1,NCI-H460及NCI-H446细胞体外生长的抑制作用。方法锥虫蓝排除法、MTT法、Hoechst33258荧光染色及DNA凝胶电泳。结果药物处理时间越长及浓度越高,细胞生长抑制作用越强,其中A549细胞株IC50为20μmol/L,敏感性最高,而NCI-H460及NCI-H446的IC50为200μmol/L,敏感性最低;荧光染色结果为所有经药物处理的肺癌细胞的染色质均产生凝集现象,在DNA凝胶电泳图中,出现梯状DNA电泳条带。结论二苯乙烯类化合物可显著抑制A549细胞的生长,且这种抑制作用表现出时间及浓度依赖性;此化合物对NCI-H460及NCI-H446细胞的生长未表现明显的抑制作用。另外。 PURPOSE To study the growth inhibitory effect of stilbenoids consisting of three components (1) (+)-viniferin, (2) miyabenol C and (3) kobophenol a kind of protein kinase C inhibitor on lung cancer cell lines A549, 95-D, SPC-A-1, NCI-H460 and NCI-H446. METHODS Trypan blue exclusion, MTT, hoechst 33258 staining and DNA agrose gel electrophoresis were used. RESULTS (1) The growth inhibitory effect of stilbenoids on lung cancer cells was strengthened gradually with the increase of treatment duration and stilbenoids concentration. The susceptibility of A549 cells to this compound was the highest, its IC50 being 20μmol/L, while that of NCI-H460 cells was the lowest with its IC50 of 200μmol/L. (2) There were chromatin comdensation and DNA ladder characteristics of apoptosis in all lung cancer cell lines treated with stilbenoid as shown by hoechst 33258 staining and DNA agrose gel electrophoresis. CONCLUSIONS The growth of A549 cells is inhibited strikingly by stilbenoids and this inhibition expressed time and concentration dependence. In addition, stilbenoids is able to induce apoptosis of lung cancer cells.
