摘要
从某柠檬酸厂分离到1株耐酸性ɑ-淀粉酶的生产菌株YC,经16SrDNA法鉴定为蜡状芽孢杆菌。通过紫外诱变及紫外-硫酸二乙酯(DES)复合诱变,选育到1株产酶稳定的正向突变株YC-UV9-D13,连续传种10代后测定其酶活仍达250.77U/mL,比野生菌株提高了4.9倍,为工业化推广奠定了良好的基础。
A wild acid-resistant α-amylase producing strain was isolated from a citric acid factory,which was identified as Bacillus cereus,using comparative 16S rDNA gene sequence analyses.A high α-amylase producing strain,YCUV9-D13,was finally bred by UV and UV+DES mutagenetic treatments,whose α-amylase activity was 250.77 U/mL after Continue propagation for ten times,and enhanced 4.9 times than the wild-type strain.These results laid a foundation for the industrial scale production of acid-resistant α-amylase.
