枯草芽孢杆菌生长培养基的优化

为获得生长活力较好,可以满足菌种水解玉米蛋白粉生产玉米肽的需要,以OD600为评价指标,采用单因素试验确定菌种生长适宜的培养基成分,即:葡萄糖2%(碳源)、大豆蛋白胨4%(氮源)、MgSO40.15%、K2HPO40.2%、KH2PO40.2%。以单因素试验菌种生长适宜的培养基成分作为二次旋转正交试验的零水平,以菌种生长14h的OD600为指标,确定培养基成分的最优组合为葡萄糖2%、大豆蛋白胨3.6%、MgSO40.16%、K2HPO40.28%、KH2PO40.28%。在此条件下菌种培养14h,其OD600由0.900上升到1.869,菌株生长量提高了1倍。

In order to gain a better strain with more stronger growth activation,to meet the requirement of the cornpeptide production from zein by enzymatic hydrolysis,Taking OD600 as evaluative index,optimum ingredient of medium for the growth of strain was determined by single factor experiment as follows：carbon source is glucose,nitrogen source is soybean peptone,optimum concentration of carbon source 2%,optimum concentration of nitrogen source 4%,MgSO4 0.15%,K2HPO 40.2%,KH2PO40.2%.then making the results as zero level of two revolving orthogonal experiment,taking the OD600 of strain culture at 14h as index,to systematically determine the optimal combination：glucose2%,soybean peptone 3.6%,MgSO4 0.16%,K2HPO4 0.28%,KH2PO4 0.28%.At this condiction,the OD600 of strain culture at 14h was ineresed from 0.900 to 1.869 the stain yield raised one time.
