吸附结合超滤技术制备高活性菠萝蛋白酶的工艺研究 被引量:1

Studies on the Technology of High Activity Bromelain Prepared by Adsorption Combined with Ultrafiltration Technique

摘要 以菠萝皮渣为原料,研究纳米TiO2吸附结合超滤、冷冻干燥技术制备菠萝蛋白酶粉的方法。结果表明:在温度15℃条件下,以粗酶液质量3.5%的纳米TiO2吸附30min,酶吸附率达99.1%;采用0.4mol/L、pH6.0的柠檬酸缓冲液洗脱,将洗脱液用截留分子质量50kD的超滤膜超滤,流出液用截留分子质量10kD的超滤膜超滤,浓缩液经冷冻干燥得到比活力为2.8×106U/g的菠萝蛋白酶粉。 Using pineapple pomace as material,the preparetion technique of bromelain by nano-TiO2 adsorption,ultrafiltration and freeze-drying method were studied.The results showed that when the addition level of nano-TiO2 was 3.5%,in the adsorption condition of 15 ℃ for 30 min,the adsorption rate of bromelain reached 99.1%.The adsorbed bromelain could be well eluted by 0.4 mol/L citrate buffer solution(pH6.0).Ultrafiltration was applied to the elution with a membrane of 50 kD.The outlet solution was collected and re-filtrated by a membrane of 10 kD,the concentrated solution was freeze-dried into bromelain powder with the enzyme activities 2.8×10^6 U/g.
