摘要
目的:了解nm23在骨肉瘤的表达及其与Ki67、早期转移的关系。方法:以抗nm23多克隆抗体和抗Ki67单克隆抗体对39例骨肉瘤石蜡组织切片进行免疫组化染色。结果:nm23阳性细胞数>25%的骨肉瘤19例,占总数的487%。转移组中nm23高表达病例的比例显著高于非转移组,但nm23的表达与病理分型和分化无关。反映肿瘤增殖因子Ki67与nm23表达呈正相关,并与骨肉瘤病理分化有关,但与转移无关。结论:本研究并未显示nm23在骨肉瘤对转移的抑制作用,但是,结合应用nm23和Ki67有可能对骨肉瘤增殖和转移潜能的评价提供有意义的信息。
Objectives:The expression of nm23 as a tumor metastasis suppressor in osteosarcomas remains unknown.The research aimed at detecting nm23 expression in osteosarcoma,analyzing the correlation between nm23 expression and early metastasis and studying the relation of nm23 expression with tumor differentiation and cell proliferation,as defined by Ki 67 labeling.Methods:nm23 oncoprotein expression was measured by immunohistochemical staining using formalin fixed,paraffin embedded sections of 39 surgically resected osteosarcomas.The pathologic types included:Osteoblastic 13 cases,Chondroblastic 9,Fibroblastic 8 and others 9.The tumors were graded as well differentiated (9 cases) and poorly differentiated (30 cases).11 cases had lung metastasis within 1 year after the operation.Results:The cases with more than 25% positive immunoreactive cells of nm23 protein were detected in 48 7% of the total cases.The cases with high nm23 expression appeared to have a trend to get early metastasis.nm23 expression was significantly correlated with ki 67 expression,which was associated with differentiation of the tumor cells.No relationships between nm23 and pathologic types,tumor differentiation,tumor sizes,history were observed.Conclusion:The role of nm23 as a tumor metastasis suppressor in osteosarcomas was less prominent.Both nm23 and ki 67 might help to predict the possibility of early metastasis in osteosarcomas.
