摘要
研究用蛋白酶消化蛋白提取完整肽聚糖的工艺,并以提取率、上清液中游离氨基氮、肽聚糖中磷含量为检测指标,应用正交试验设计优化消化酶的种类、酶的浓度及提取时间3个因素。结果表明,胰酶和碱性蛋白酶在活力单位为3500NFU,处理12h达到最佳效果,所得肽聚糖为白色絮状物质,提取率为7.3%,上清液中释放的游离氨基氮最多为686.9μg/mL,肽聚糖磷的含量0.95mg/g。
In this study, whole peptidoglycan were isolated from bifidobacterium infantis through double-enzyme hydrolyzed, and thedetection index contained extraction ratio, free amino nitrogen in supernatant, phosphorus content of peptidoglycan, the quadrature of experimental design optimized the type of enzyme, enzyme concentration and extraction time. The results showed that the alkaline protease and trypsin in 3 500 NFU, dealing with 12 h were the best, peptidoglycan is white flocculent material, extraction ratio was 7.3%, free amino nitrogen in supernatant uped to 686.9 μg/mL, peptidoglycan phosphorus was 0.95 mg/g.
