桃核承气改良方对家兔血管内皮损伤后PDGF-B和VEGF影响的实验研究 被引量:3

摘要 目的:研究中药桃核承气改良方对家兔血管内皮损伤后PDGF-B(血小板源生长因子B蛋白)和VEGF(血管内皮生长因子)的影响。方法:健康新西兰大耳白兔40只,体重2~2.5kg。将动物分模型组(n=10)、中药组(n=10)、西药组(n=10)、空白组(n=10),模型组、中药组、西药组高酯饲料喂养4周后行腹主动脉内膜剥脱术造模。术后模型组、中药组、西药组正常饲料喂养,中药组予桃核承气改良方灌胃4周,西药组予舒降之、阿司匹林灌胃4周,模型组、对照组予生理盐水灌胃4周。造模术4周后处死家兔,用免疫组化法观察各组对血小板源性生长因子(PDGF-B)表达的影响;采用半定量RT-PCR法测定血管中VEGFmRNA半定量,采用WesternBlot(免疫印迹)印迹杂交法结果量化分析。结果:与模型组比较,中药组内膜/中膜厚度比、内膜/中膜面积比显著减少(P<0.05);中药组使血管壁PDGF-B含量下降;模型组VEGFmRNA半定量、蛋白表达较对照组明显升高(P<0.05);西药组VEGFmRNA半定量、蛋白表达较模型组明显降低(P<0.05);中药组VEGFmR-NA半定量、蛋白表达较模型组明显降低(P<0.05),中药组与西药组之间无明显差异(P>0.05)。结论:桃核承气改良方对血管内膜增殖早期血小板源生长因子B蛋白(PDGF-B)有拮抗作用,对血管内皮损伤有一定的保护作用;抑制血管内皮生长因子(VEGF)从而预防冠心病经皮冠状动脉介入治疗技术术后冠脉再狭窄。 Objective : To study the improvement of Chinese medicine Taohe Chengqi on vascular endothelial injury in rabbits after PDGF - B ( platelet - derived growth factor B protein) and VEGF ( vascular endothelial growth factor) affected. Methods : New Zealand 40 rabbits, weighing 2 ~ 2.5kg. Animals divided into model group ( n = 10), traditional Chinese medicine group (n =10), chemical medicine group (n = 10), the control group (n = 10), model group, the Chinese Medicines Board, chemical medicine group fed the high - ester 4 weeks after abdominal aortic endarterectomy model. After the model group, the Chinese Medicines Board, chemical drugs and normal diet group, the Chinese Medicines Board to improve the side Taohe Chengqi gavage for 4 weeks, the chemical group to the drug Zocor, aspirin orally for 4 weeks, model group, control group to the saline gavage for 4 weeks. Surgery model in rabbits were sacrificed after 4 weeks from the abdominal aorta artery pathological changes observed, measured intima, media thickness and area, by im- munohistochemical method to observe the group of platelet - derived growth factor ( PDGF - B) expression impact ; using semi - quantitative RT - PCR determination of VEGF mRNA in blood vessels semi - quantitative, using Western Blot (immunoblot) blot hybridization results of quantitative analysis. Results: Compared with model group, the Chinese Medicines Board intima / media thickness ratio and intima / media area ratio significantly reduced (P 〈 0.05) ; the Chinese Medicines Board to make the vessel wall decreased PDGF - B ; model group, VEGF mRNA half - quantitative, protein expression significantly higher than the control group ( P 〈 0.05 ) ; chemical drug - group semi - quantitative VEGF mR- NA and protein expression was significantly lower than the model ( P 〈 0.05 ) ; the Chinese Medicines Board semi - quan- titative VEGF mRNA and protein expression in model group was significant lower ( P 〈 0.05 ) , traditional Chinese medi- cine and chemical medicine group was no significant difference between groups ( P 〉 0. 05 ). Conclusion : The improved nuclear Dachengqi peach on vascular intimal proliferation of early platelet - derived growth factor B protein ( PDGF - B) has antagonistic effects on vascular endothelial injury have some protective effect; prevention and treatment of vascular endothelial growth factor and thus the prevention of percutaneous coronary heart disease after coronary intervention treatment of coronary restenosis.
