不同浓度人参皂苷RG3对高转移卵巢癌细胞集落形成及细胞形态的影响 被引量:3

Influence of ginsenoside Rg3 on cell colony-forming efficiency and morphocytology of human high-metastasis ovarian cancer cell line H0-8910PM in vitro

摘要 目的:观察人参皂苷( Rg3)对高转移人卵巢癌细胞系H0-8910PM的细胞集落形成和细胞形态的影响。方法:采用本院自建的高转移人卵巢癌细胞系H0-8910PM进行实验,随机分成5组:①空白对照组:只加新鲜的全培养液;②低浓度 Rg3组:10μg/mL Rg3培养液;③中浓度 Rg3组:20μg/mL Rg3培养液;④中高浓度 Rg3组:30μg/mL Rg3培养液;⑤高浓度 Rg3组:40μg/mL Rg3培养液。观察不同浓度 Rg3分别对细胞集落形成率和细胞形态的影响。结果:经单因素分析发现5组细胞随着 Rg3浓度的升高,细胞集落形成率逐渐而下降,差异有统计学意义(F=176.362,P=0.000)。多组变量随机区组分析示:空白对照组与任何浓度 Rg3组比较差异均有统计学意义(P=0.000,0.000,0.000,0.000)。低浓度组与中高浓度 Rg3两组细胞集落形成率结果相近(8.3%vs6.9%),差异无统计学意义(P=0.133)。中高浓度组与高浓度 Rg3两组细胞集落形成率均很低(2.5%Vs0.78%),差异无统计学意义(P=0.057)。低浓度组、中高浓度组分别与中高浓度组、高浓度组比较,差异均有统计学意义(P=0.000,0.000,0.000,0.000)。中浓度以上 Rg3可引起H0-8910PM的细胞形态结构损伤。中高浓度及高浓度 Rg3在微观结构上能造成更严重的细胞形态结构损伤。结论: Rg3对高转移人卵巢癌H0-8910PM细胞能有效减少细胞集落的形成。中高、高浓度的 Rg3(30μg/mL以上)尤其可以产生较好的抑制肿瘤细胞生长作用,并呈一定的剂量依赖性,且对细胞形态结构破坏上产生更重的损伤。 Objective : To observe the effect of cell colony - forming efficiency and morphocytology with Ginsenoside Rg3 on high - metastasis human ovarian cancer cell line H0 - 8910PM. Methods : The cell line were divided into 5 groups randomly: (1)blank control group: only adding fresh complete culture medium; (2)low consistency Rg3 group: 10μg/mL Rg3 medium; (3)middle consistency Rg3 group : 20μg/mL Rg3 medium ; (4)middle - high consistency Rg3 group : 30μ g/mL Rg3 medium; (4)high consistency Rg3 group: 40μg/mL Rg3 medium. In this study cell colony -forming effciency and morphocytology changement were used to investigate the effects of cell proliferation suppression. Results : Univariance analysis showed in these 5 groups the higher concentration of Rg3 , the lower the cell colony - forming effciency ( F = 176.362, P =0.000). Multiple comparisons showed any group of Rg3 had significant different with control group( P = 0.000). The cell colony - forming effciency of middle - high consistency and high consistency Rg3 groups had no significant different(8.3% vs 6.9%, P = 0. 133) ; also with low consistency and middle consistency Rg3 group(2.5% vs 0. 78%, P =0.057). low consistency or middle consistency Rg3 group had significant different with middle -high or high consistency Rg3 group respectively(all P =0.000 ). Above 10μg/mL Rg3 medium could cause H0 -8910PM cell morphology damage, but groups with consistency more than 30tzg/mL could make more severe damages in microstructure and cellular morphology. Conclusions : Rg3 alone has shown the inhibition of cell colony - forming efficiency of human ovarian cancer cell line H0 - 8910PM. Rg3 also has obvious dose - dependently anti - tumor effect in groups with consistency more than 30μg/mL Rg3. Middle - high and high consistency Rg3 groups may be more appropriate strategies about both inhibition of proliferation and morphocytology damage.
