地骨皮提取液对2型糖尿病肥胖大鼠糖脂代谢影响的实验研究

Improvement of Cortex Lycii Radicis Extracts on Glucolipids Metabolism in Fatty Rats with Type 2 Diabetes Mellitus

目的:观察地骨皮提取液对2型糖尿病肥胖大鼠(T2DM)糖脂代谢及胰岛素抵抗(IR)的影响,并对醇提、水提两种提取液进行比较。方法:采用尾静脉注射小剂量链脲佐菌素(STZ)加高脂饮食饲养的方法建立T2DM肥胖大鼠IR模型,设立空白对照组、模型对照组、地骨皮醇提、水提高/低剂量组,给药7周后观察对大鼠体重、血糖、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、胰岛素敏感指数(ISI)的影响,及肝脏的病理变化。结果:与模型组相比,地骨皮提取液能降低T2DM肥胖大鼠的体重、血糖、TG、TC、LDL-C,差异有显著性(P<0.01),能明显改善ISI(P<0.01);与空白对照组相比无明显差异。肝脏病理显示能明显降低高脂饮食对肝脏的损害,保护肝脏细胞。结论:地骨皮提取液可在一定程度上降低血糖,调节血脂代谢,缓解肝脂肪变性程度,改善T2DM肥胖大鼠的IR状态;而且醇提液优于水提。

To evaluate effects of ethanol and aqueous extracts from Cortex Lycii Radicis （CLR） on Insulin Resistance and lipid metabolize in fatty rats with type 2 diabetes, induced by feeding high fat diet for 3 weeks after injection with Streptozotocin （STZ）. Diabetic rats treated with ethanol or aqueous extracts of Cortex Lycii Radicis at 15 and 30 mg/kg dosage for 7 weeks, were found that body weights, concentration of serum glucose, triglyeeride （TG）, total cholesterol （TC） , and low density lipoprotein - cholesterol （ LDL - C ） , alanine anainotransferase （ALT） , aspartate aminotransferase （AST） decreased obviously while insulin -sensitivity index （ISI） improved significantly compared with the model group. In addition, histopathologie examination showed the decreased hepatoeytes edema with little small - drop fat and partly hepatoeytes ballooning degeneration in all Cortex Lycii Radicis groups. According to the whole results, ethanol extract from the herb seemed to be more effective than aqueous extraet. These findings implicated that extracts of Cortex Ly- eli Radieis could improve insulin resistance, resulted in elevating insulin sensitivity, adjusting glucose and lipid metabolism and ameliorating liver function, especially protecting liver against the lipoid degeneration.
