5-氮杂胞苷对IL-6启动子DNA甲基化的影响

摘要 目的:探讨DNA甲基化抑制剂对健康人外周血T淋巴细胞IL-6启动子的DNA甲基化水平以及IIL-6表达的影响。方法:用DNA的亚硫酸氢钠修饰、巢式PCR和PCR产物直接测序检测10例甲基化抑制组(5-氮杂胞苷抑制甲基化的健康人T细胞)和对照组(未经5-氮杂胞苷抑制甲基化的健康人T细胞)的外周血T细胞启动子的甲基化水平,IL-6转录水平则用RT-PCR方法。结果:甲基化抑制组的T细胞IL-6启动子DNA甲基化水平较对照组明显降低,而IL-6表达较对照组明显增加。结论:5-氮杂胞苷确实可以抑制健康人外周血T淋巴细胞IL-6启动子的DNA甲基化水平并促进IL-6的表达。 Objective:To investigate the role of DNA methylation in the expression of interleu- kin - 6 ( IL - 6) in T cells. Methods : The methylation status of IL - 6 of T cells from 10 healthy control activated in vitro in the presence or absence of 5 - azaCytidine (5 - azaC) were character- ized using DNA bisulfate modification followed nested - PCR and direct sequencing. IL -6 mRNA levels from these cells were detected with RT - PCR. Results: The methylation level of IL - 6 pro- moter decreased and the levels of IL - 6 mRNA transcripts were elevated in 5 - azaC treated T cells as compared with control. Conclusion : Administration of 5 - azaC is able to inhibit the meth- ylation level of IL -6 promoter and increase level of its transcription.
