不同CpG ODN对大鼠骨髓间充质干细胞增殖作用的研究 被引量:6

摘要 目的:筛选对大鼠骨髓间充质干细胞增殖具有影响作用的CpG ODN。方法:体外全骨髓贴壁法提取并纯化大鼠骨髓间充质干细胞,传至第3代孵育12 h至完全贴壁,双盲法加入不同CpG ODN共孵育48 h,MTT比色法检测,不同大鼠重复实验3次,筛选促进大鼠骨髓间充质干细胞增殖的CpG ODN。结果:与PBS溶媒对照组相比,有4条CpG ODN对不同大鼠BMSCs均具有显著促增殖作用,MTT结果具有统计学意义(P<0.05)。结论:4条CpG ODN具有显著促进大鼠BMSCs增殖的作用,将在下一步的实验中探讨它们对BMSCs分化功能的作用。 Objective: To screen the optimal CpG ODN for inducing the proliferation of rat bone marrow mesenchymal stem cells (BMSCs). Methods: The third generation of BMSCs that were separated and purified from rat were plated 12 h before CpG ODNs was added. MTT assay was used to detect cell proliferation after 48h coculture. The above experiments were repeated for 3 times. Results: The mean OD values of four CpG ODNs' groups were significantly higher than those in PBS solvent control group and other CpG ODNs groups (P〈0.05 vs all of others). Conclusion: The four CpG ODN sequences that significantly promote the proliferation of rat BMSCs will be used for the next experiment to explore their effects on the functional differentiation of MSCs.
