脐带基质干细胞移植在卵巢早衰模型小鼠体内的分布 被引量:11

摘要 目的观察人脐带基质干细胞(umbilical cord-matrix stem cells,UC-MSCs)移植后在卵巢早衰(premature ovarian failure,POF)小鼠模型中的分布与生长情况。方法用环磷酰胺(cyclophosphamide,CPA)和白消安(busulfan,BUS)处理,建立小鼠POF模型;分离扩增UC-MSCs,采用腺病毒介导Ad-GFP转染方式对其进行标记;将GFP阳性细胞移植入POF小鼠体内,分别于移植后1、10、20 d留取组织,通过激光共聚焦显微镜观察UC-MSCs在小鼠卵巢及其他组织中的定位与生长情况。结果腺病毒介导Ad-GFP转染UC-MSCs效率达95%左右。POF小鼠经尾静脉接受UC-MSCs移植后1 d,卵巢可见GFP阳性细胞,移植后10 d及20 d的卵巢GFP阳性细胞明显增加,而小鼠骨髓、大脑、肝脏、肾脏、胰腺等组织无明显GFP阳性细胞存在。结论人UC-MSCs经尾静脉移植可向POF小鼠卵巢归巢,并能在局部存活生长,提示其可能具有卵巢损伤修复的作用。 Objective To observe the localization cells (UC-MSCs) in mouse premature ovarian failure (POF) of transplanted human umbilical cord-matrix stem model. Methods POF animal model was established by cyclophosphamide (CPA) and busulfan (BUS) chemotherapy. After UC-MSCs were isolated and proliferated from UC, recombinant Ad-enovirus-green fluorescent protein (Ad-GFP) was transfected in them via Ad-enovirus. The GFP-transfected UC-MSCs (2 × 10^6 to 3 × 10^6) were transplanted into POF mice through caudal vein, and tissues from POF mice were obtained for frozen section at 1, 10 and 20 d after transplantation. GFP-positive cells in the ovary, brain, liver, kidney and pancreas were determined by laser confocal microscopy. Results Transfection efficiency of Ad-GFP by Ad-enovirus was about 95%. After transplantation, GFP- positive cells were found in the harmed ovaries and more GFP-positive cells were found at 10 and 20 d after transplantation than those at 1 d after transplantation. However, no GFP-positive cells were found in the bone marrow, brain, liver, kidney and pancreas. Conclusion Human UC-MSCs can home and survive in the ovaries of POF mouse model. It suggests that UC-MSCs maybe have the potential for POF therapy.
