摘要 以废蚕丝为原料,研究了蚕丝固定β-葡萄糖苷酶的方法。采用共价法和包埋法将酶固定在丝素蛋白膜上,操作简便易行,制造的酶膜稳定,机械性能好,热稳定性提高,70℃保存1小时活力几乎不降低,同时pH值稳定性也有所提高。该方法对果酒增香具有实用价值。 The β glucosidase is immobilized on silk fibroin membrane by entrapped method and covalent crosslinking method with glutaraldehyde, with the activity recovery of the immobilized enzyme being 80% and 20%-30%, respectively. The enzymatic properties are investigated of covalent glucosidase silk membrane and of free enzmye. It is demonstrated that the immobilized enzyme shows higher pH and thermal stability. Optimum pH of the activity of the immobilized enzyme is shifted from 4.5 to the value around neutrality. The value of kmapp of immobilized β glucosidase is less than approximately one fifth of the free enzyme. In addition, the enzyme membrane has good storage stability and operating stability.
