摘要
【目的】研究5-氟尿嘧啶和奥沙利铂分别诱导结肠癌细胞株SW620的细胞凋亡的作用效果。【方法】用80μg/ml5-氟尿嘧啶和25μg/ml奥沙利铂分别作用SW620细胞8h,并应用AnnexinV-FITC/PI双标记流式细胞术对细胞凋亡进行检测,观察凋亡细胞的比例。【结果】80μg/ml5-氟尿嘧啶处理8h组与空白对照组比较,凋亡细胞比例上升约11.9%(P<0.01);25μg/ml奥沙利铂处理8小时组与空白对照组比较,凋亡细胞比例上升约21.4%(P<0.01)。【结论】在特定条件下,5-氟尿嘧啶和奥沙利铂均能明显诱导结肠癌细胞株SW620的细胞凋亡;奥沙利铂对结肠癌细胞的诱导凋亡作用要强于5-氟尿嘧啶。
[Objective] Separately observe the effect of apoptosis being induced in colon cancer cell line SW620 by 5-FU and oxaliplatin under specific condition. [ Methods ] SW620 cells were separately treated with 80 m g/ml 5-FU and 25 μ g/ml oxaliplatin for 8 hours. And analyzed apoptosis by AnnexinV-FITC/PI double staining flow cytometry. Observe the proportion of apoptosis cells. [Results] SW620 cell line was detected by AnnexinV-FITC/PI double staining flow cytometry. Compared with the blank group, the proportion of apoptosis cells of the group treated with 80 μ g/ml 5-FU for 8 hours increased approximately 11.9% ( P 〈 0.01). Compared with the blank group, the proportion of apoptosis cells of the group treated with 25 μg/ml oxaliplatin for 8 hours increased approximately 21.4% ( P 〈 0.01). [Conclusion] SW620 cell line apoptosis can be obvious induced by 5-FU and oxaliplatin under specific condition. Oxaliplatin can do more apoptosis induced than 5-FU to colon cancer cells.
