豚鼠真菌性角膜炎模型的建立及病理观察 被引量:3

摘要 目的探讨理想的真菌性角膜炎动物模型的制作方法,并观察其病理学改变。方法豚鼠25只(50眼)制作真菌性角膜炎动物模型。分别于接种后2d、4d、7d、14d过量乙醚麻醉处死豚鼠,取角膜,光镜下观察组织学改变及真菌孢子和菌丝。结果接种真菌2d后,可见角膜全层水肿,溃疡形成,干燥致密,粗糙不平,稍隆起。HE染色显示角膜上皮层可见散在孢子及菌丝,且菌丝向基质层长入。随病情进展,溃疡部位角膜上皮及浅基质修复,基质内较多成纤维细胞,细胞走行与基质纤维平行,菌丝平行或斜行于角膜板层生长,有分隔,圆形孢子散在生长,部分标本前房可见菌丝和孢子。结论利用豚鼠采用剖除角膜上皮,眼睑缝合,菌液滴眼的方法可以成功建立真菌性角膜炎的动物模型。 Objective To investigate the ideal way to make animal models with fungal keratitis and to observe its histopathological changes.Methods Twenty five(50 eyes) cavia cobaya were selected to establish fungal keratitis models,and were sacrificed with overdose etherization at 2 days,4 days,7 days and 14 days after inoculation.The corneas were taken,histological changes,fungal spores and hyphae were observed under light microscope.Results After inoculating fungus for 2 days,there were corneal edema,ulceration,dryness and tightness,rough,uneven and swelling.Pathological staining showed that scattered fungal spores and hyphae were seen at corneal epithelium,and hyphae grew into corneal stroma.With the progress of pathogenetic condition,light corneal stroma and corneal epithelium were recovered at ulcer,more fibroblasts were seen at corneal stroma,cell lining was parallel with matrix fiber,hyphae was parallel or oblique to corneal layer.There was separation,round spores grew in scatter,way and hyphae and spores were seen at anterior chamber in part specimen.Conclusion With eliminating corneal epithelium,eyelid suturing and fungus fluid eyedropping,cavia cobaya can be used to establish animal models with fungal keratitis successfully.
