三种去除人巨噬细胞迁移抑制因子(HuMIF)重组蛋白内毒素方法的比较 被引量:1

摘要 运用3种方法(C8反相柱层析.复性法、多步层析法和本实验室改进的c8反相柱层析.非复性法)去除重组HuMIF蛋白的内毒素,比较其所得蛋白内毒素含量、酶活性以及蛋白回收率等,评价这3种去内毒素方法的优劣。结果表明,c8反相柱层析.复性法蛋白回收率很低,酶活性低但内毒素去除效果好;多步层析法蛋白回收率高,酶活性高但内毒素去除效果稍差;而本实验室改进的c8反相柱层析.非复性法蛋白回收率稍差,酶活性较好且内毒素去除效果不错。由于c8反相柱层析一非复性法内毒素去除方法简单有效,适合实验室范围内推广。 Three different protein endotoxin removal methods including C8 reversed-phase column chromatography-renaturation, Multi-step chromatography and an improved C8 reversed-phase column chromatography-non-renaturation were compared in present paper. The evaluation items included endotoxin protein content, enzyme activity and protein recovery. The results showed that the C8 reversed-phase column chromatography-renaturation may get very low protein recovery rate, low protein activity but good endotoxin removal efficiency; multi-step chromatography may get high protein recovery rate, high protein activity and less endotoxin removal efficiency; while reversed-phase column chromatography-non-renaturation may get low protein recovery rate, relatively high protein activity and good endotoxin removal efficiency. As a result, it seems that C8 reversed-phase column chromatography-non- renaturation method might be suitable for laboratory application and dissemination for its simple handling and effectiveness.
