罗勒胶囊对大鼠移植性肝癌TACE后HIF-1α和Bcl-2表达的影响

Effects of Luole capsule on expressions of HIF-1α and Bcl-2 for transplanted liver cancer in rats after TACE

目的:观察罗勒胶囊对大鼠移植性肝癌行经导管肝动脉化疗栓塞术(TACE)后组织中乏氧诱导因子(α)(HIF-1α)和凋亡抑制基因Bcl-2表达的影响,探讨该药对肝肿瘤乏氧微环境的作用。方法:建立大鼠移植性肝癌模型后,随机分为5组。对TACE+生理盐水组和TACE+罗勒胶囊大、中、小剂量组大鼠进行TACE,并分别给予生理盐水和大、中、小剂量罗勒胶囊灌胃,对非TACE对照组大鼠只进行开腹操作,不进行TACE。10d后取出移植性肝癌病理标本,免疫组化法检测肿瘤组织中HIF-1α和Bcl-2表达情况。结果:行TACE的4组大鼠瘤重与非TACE组比较均明显减轻(P<0.05),TACE+罗勒胶囊大剂量组瘤重较TACE+生理盐水组显著减轻(P<0.05);HIF-1α和Bcl-2阳性表达率方面:TACE+生理盐水组与非TACE组比较显著升高(P<0.01),TACE+罗勒胶囊大、中剂量组较TACE+生理盐水组显著降低(P<0.05)。结论:罗勒胶囊能使肿瘤组织中HIF-1α和Bcl-2表达降低,改善肝肿瘤局部的乏氧微环境并促进细胞凋亡,抑制大鼠移植性肝癌TACE术后肿瘤的复发。

Objectives：To study effects of Luole capsule on expressions of HIF-1α and Bcl-2 for transplanted liver cancer in rats after transcatheter arterial chemoembolization（TACE）,and discuss the significance of the capsule on hypoxia Methods：The liver tumor models were established and were randomly divided into five groups ：without TACE group,no medicine to use ;TACE＋ Saline （B）group,were given Saline after TACE,TACE＋large dose Luole capsule （C）group, TACE＋mid-dose Luole capsule （D）group, TACE＋mini-dose Luole capsule （E）group, carrying out different dose Luole capsule after TACE. After ten days,the tumors were taken out. To demonstrate the expression rates of HIF-1α and Bcl-2 with immunohistochemistry. Results：The tumor weights of the groups after TACE were significantly reduced than A group（P〈0.05）,the tumor weight of C group was reduced than B group（P〈0.05）. The expression rates of HIF-1α and Bcl-2 of B group were higher than A group（P〈0.01）;the expression rate of HIF-1α and Bcl-2 of C group and D group were lower than B group（P〈0.05）;the expression rate of HIF-1α and Bcl-2 between C group and D group were different（P〈0.05）. Conclusion：Luole capsule could depress the expressions of HIF-1α and Bcl-2,could obviously improve local hypoxia micro-environment in liver tumor rats and inhibit rat liver transplant tumor recurrence after TACE.
