紫杉醇诱导人体外培养滑膜细胞凋亡 被引量:1

Taxol inducing apoptosis of human cultured synoviocyte in vitro

摘要 目的研究紫杉醇对类风湿关节炎(RA)的作用。方法用光镜、电镜、共聚焦显微镜、流式细胞仪及DNA凝胶电泳等方法检测了紫杉醇诱导体外培养的9例RA与5例骨关节炎(OA)及4例正常对照(骨折)患者关节滑膜细胞凋亡(Apo)情况。结果紫杉醇对3种体外培养的滑膜细胞均可诱导出现凋亡,其中RA组最高,可达6704%±1120%。OA组为3828%±440%,正常对照组为1516%±875%。RA组凋亡细胞百分率显著高于OA组(P<0.01)和正常对照组(P<0.01),在一定剂量范围内具有时间和浓度依赖性。结论紫杉醇可能具有临床治疗RA的潜在性价值。 Objective To study the role of taxol in rheumatiod arthritis (RA). Methods Apoptosis of joint synoviocytes induced by taxol from 9 cases of RA, 5 of osteoarthritis (OA) and 4 normal controls (NC, cases of acute trauma) was detected in vitro with light microscope, electron microscope, confocal laser scanning micrograph, flow cytometric analysis and DNA eletrophoresis. Results Apoptosis could be induced by taxol in all of the synoviocytes cultured in vitro. Of the three groups of RA, OA, NC, the rate of apoptotic cells was in RA 67.04%+11.20% and those of 38.28%+4.40% and NC 15.16%+8.75% were lower. The rate of the apoptotic cells of RA was significantly higher than those of OA ( P <0.01) and NC ( P <0.01). There was dependence of time and dose within a certain range. Conclusion Taxol is of clinical potential value for the treatment of RA. To induce synoviocyte apoptosis in RA may provide a new method for finding new medicines to treat RA.
