摘要
采用二巯基乙醇直接还原法将Tc99m放射性核素标记抗人前列腺癌单克隆抗体YDPC,标记物经SephadexG50柱,获得纯化的标记抗体YDPC99mTc,薄层层析检测其放射化学纯度为97%。YDPC99mTc经尾静脉注射入荷人前列腺癌裸鼠体内,24小时后行γ照相,可以得到前列腺癌的特异显像,而同一裸鼠体内的人直肠癌移植瘤没有显像。结论:YDPC99mTc放射免疫显像具有前列腺癌特异性。
Anti human prostate cancer monoclonal antibody YDPC was labeled with technetium 99m by direct 2 mercaptoethanol reduction method. The labeled antibody YDPC 99m Tc was purified through Sephadex G 50 column chromatography. The radiochemical purity was 97%. YDPC 99m Tc was injected through tail vein of nude mice xenographed with human prostate cancer. The animal was scintigraphed with γ camera 24 hours later to get a clear and specific image of the tumor. These results indicate that the radioimmunoscintigraphy of prostate cancer is specific.
