摘要
为探讨一氧化氮(NO)在人体膀胱的生理和病理改变中的作用,应用免疫组化法和Western印迹法对人体正常膀胱(10例),前列腺增生症者稳定膀胱(9例)和不稳定膀胱(12例)逼尿肌组织中的一氧化氮合酶(NOS)进行检测。结果显示:膀胱逼尿肌组织中有NOS表达,神经节、神经元、神经束和神经纤维皆呈阳性反应,但各类膀胱间的表达量无显著性差异(P>0.05)。提示NO介导的神经参与调节膀胱逼尿肌的松弛。
Expression of NOS in bladder detrusors was studied in 12 BPH patients with unstable bladder,9 BPH patients with stable bladder and in 12 male patients with bladder cancer but with no mechnical obstruction or detrusor instability.All the patients had undergone a bladder surgery and during the operation 2 small pieces of detrusor were cut from the edge of the incision on the anterior bladder wall.One sample each was used for immunohistochemical study and the other for western blot study.Density of the protein bands was analysed with image analyse system.The result revealed that there were NOS like immunoreactive nerves in the bladder detrusor with nervous constitutive NOS positively expressed.However,there was no significant quantitative difference of ncNOS among different groups of patients ( P >0.05).It was concluded that the NO mediated nerves do modulate the relaxation of bladder detrusor,but the modulation is too weak and do not play the main role in the pathological mechanism of an unstable bladder.
