摘要
以南极海冰细菌作为实验材料,用刚果红染色和发酵培养法从64株细菌中筛选得到产纤维素酶菌株Pseudoalteromonassp.545。经生长条件优化,该菌以麦芽糖为碳源,蛋白胨为氮源,初始生长pH为8.0,10℃培养96h时产酶最高。对菌株粗酶液进行酶学性质初步研究,此酶在pH9.0、35℃下反应40min时酶活最高。另外,该菌株所产纤维素酶在0~60℃范围内均有活性,属于低温酶。
Primarily screened by cellulose-congo red plate and then screened by shake flask fermentation, an effective cellulose-producing bacterium named Pseudoalteromonas sp. 545 was isolated from 64 antarctic bacteria strains. With maltose as carbon source, peptone used as nitrogen source, when 545 cultured at pH8.0 for 96 h under 10 ℃, it could produce the highest enzyme activity. Through the preliminary study of enzymatic characterization, the results showed the optimum pH of crude cellulose was 9.0, the optimum temperature 35 ℃ at 40 min. In addition, the cellulose kept its activity from 0 ℃ to 60 ℃, and belonged to low-temperature enzyme.
