摘要
用纸电泳法分离检测果糖二磷酸钠中有关物质6磷酸果糖及果糖,分离效果好,方法简便快速,检出灵敏度高。以枸橼酸盐缓冲液(pH30)为电泳缓冲液,采用中速薄型定量层析滤纸作电泳滤纸,电压梯度18V/cm,电泳80min,用盐酸间苯二胺乙醇液显色。以果糖二磷酸钠泳动距离为100,6磷酸果糖和果糖的相对迁移率(n=12)分别为074±006、012±003。最低检出量分别为果糖二磷酸钠2μg,6磷酸果糖5μg,果糖025μg。
A new and sensitive paper electrophoresis method for the analysis of the related substances fructose and fructose 6 phosphate in fructose 1,6 diphosphoric acid trisodium is described. The voltage was adjusted to about 18 V/cm of the electrophoresis paper and electrophoresis was carried out for 80 min.The buffer solution for paper electrophoresis was citrate buffer pH 3 0.Colour developing agent was m-phenylenediamine hydrochloride solution in ethanol. Taking the migration rate of fructose 1,6 diphosphoric acid trisodium salt was 1 00,the relative migration rates(n=12)were 0 74±0 06 and 0 12±0 03 for fructose 6 phosphate and fructose,respectively.The limit of detection was 0 25 μg for fructose,5 μg for fructose 6 phosphate and 3 μg for fructose 1,6 diphosphoric acid trisodium.
