能全素对梗阻性黄疸幼鼠肝脏ET mRNA表达的影响 被引量:2

Effects of Nutrison on Liver Endothelin mRNA Expression in Growing Rats with Obstructive Jaundice

摘要 目的研究梗阻性黄疸对幼鼠肝脏内皮素(ET)mRNA表达的影响和能全素对肝脏ET mRNA表达的调节,探讨临床应用肠内营养改善梗阻性黄疸患者肝损害的理论依据。方法将40只Wistar幼鼠随机分为正常对照组、假手术组、梗阻性黄疸组(以下简称OJ组)、OJ+能全素(N)组,每组10只。模型制作后给予肠内营养,检测实验鼠肝功能和形态学;应用RT-PCR检测肝组织ET mRNA表达,对所得数据进行统计学处理。结果①OJ+N组ALT较OJ组显著降低;②OJ组肝细胞淤胆、变性、胞浆疏松化、气球样变。OJ+N组肝细胞变性较轻。③OJ组肝脏内皮素(ET)A,B mRNA表达均显著高于对照组及OJ+N组,均P<0.05。结论梗阻性黄疸幼鼠肝脏结构和功能及肝组织ET mRNA表达显著改变。应用肠内营养的OJ幼鼠肝脏病理形态和肝功能较OJ组显著下降,可能与其减轻肝脏ET mRNA的表达有关。 Objective To investigate the relationship between hepatic tissue endothelin mRNA( ET mRNA) expression and obstructive jaundice( OJ) ,and effect of nutrison on the level of ET mRNA. Methods Forty immature male Wistar rats were randomly divided into four groups : control, sham operation, OJ, and OJ + N. After creation of experimental jaundice model, Nutrison were orally given to rats with OJ. Liver function and morphology,ET mRNA were detected. Results (1)Serum ALT value in experimental jaundice group was significantly higher than that of control, sham operation and OJ + N groups. (2)In OJ group, obvious degeneration of hepatocytes was observed, while in OJ + N group, the pathologic changes were milder than that of OJ group. (3)The ETA and ETB mRNA over-expression in OJ group were observed compared with control, OJ + N groups. Conclusion In growing rat model of experimental obstructive jaundice, hepatic morphology and liver biochemical tests altered significantly and ETA and ETB mRNA over expression were demonstrated in OJ group, enteral nutrition can improve liver function and decrease hepatic histopathology and ET A, B mRNA over-expression. The enteral nutrition is effective in preventing liver damage in obstructive jaundice. The mechanisms of the preventive effects may contribute to the reduction of expression of ETA, ETB in hepatic tissue by enferal nutrition.
