K-ras基因与c-myc基因在大肠癌中的表达及相关性研究 被引量:7

摘要 目的通过研究k-ras基因与c-myc基因在大肠癌中的表达情况,初步探讨二者在大肠癌发生、发展过程中的作用及可能的分子机制。方法采用S-P免疫组织化学方法检测80例结直肠癌组织及15例正常大肠黏膜中k-ras基因与c-myc基因的表达情况。结果大肠癌组织中k-ras阳性表达率65.0%(52/80),c-myc阳性表达率52.50%(42/80)。k-ras基因与c-myc基因在正常组织中未见表达,在大肠癌组织中呈阳性表达。随着分化程度的降低k-ras基因与c-myc基因的阳性表达率也明显增高(P<0.05);k-ras突变与年龄、性别、有无淋巴结转移无相关性。c-myc阳性表达与大肠癌的临床分期、淋巴结转移密切相关(P<0.05);k-ras基因与c-myc基因之间存在明显的相关性(P<0.05)。结论k-ras基因在大肠癌中的表达与细胞分化程度、Dukes分期有密切关系;c-myc在大肠癌中的表达与细胞分化程度、Dukes分期及有无淋巴结转移有密切关系;而k-ras基因与c-myc基因的表达与年龄、性别无明显关系。k-ras基因与c-myc基因的表达存在明显的正相关关系。 Objective To examine the expressions of k-ras and c-myc in colorectal carcinoma,and to analyze their correlation. Methods The expressions of k-ras and c-myc proteins were detected in 80 colorectal carcinoma samples and 15 normal mucosas by immunohistochemistry S-P method,and their correlation was then analyzed. Results The positive expressions of k-ras was 65. 0% (52 /80),and c-myc was 52. 50% (42 /80). k-ras and c-myc were rarely seen in normal organism,but were remarkably decreased in colorectal correlation with age and sex.The expressions of k-ras and c-myc direct correlation each other in colorectal carcinoma.
