“头穴丛刺法”对局灶性脑梗死大鼠可塑性蛋白MAP-2的影响 被引量:19

Study on the effect of cluster needling of scalp acupuncture on the plasticity protein MAP-2 in rats with focal cerebral infarction

摘要 目的:探讨"头穴丛刺法"对局灶性脑梗死大鼠神经可塑性影响的物质基础和可能机制。方法:采用线栓法制备大脑中动脉急性脑梗死大鼠模型,将132只雄性Wistar大鼠随机分为假手术组(A组)、模型组(B组)、头穴透刺组(C组)及头穴丛刺组(D组)。C组采用针刺"百会"透"曲鬓"穴;D组采用"头穴丛刺"("百会"及"百会"左右侧各旁开4 mm处)。均快速捻转1 min后留针30 min,每日1次,6 d为一疗程,共治疗4个疗程。A组与B组不予治疗。采用Bederson评分评价神经功能变化;免疫组织化学染色(S-P)法观察缺血半暗带微管相关蛋白2(MAP-2)阳性信号的表达。结果:7 d时,D组与B组比较,大鼠神经功能评分降低(P<0.05);14 d、28 d时,C组、D组分别与B组比较,神经功能评分均降低(均P<0.05);28 d时D组与C组比较,D组神经功能评分降低(P<0.05)。7 d、14 d、28 d时,D组、C组与B组比较,缺血皮层MAP-2阳性信号的表达均上升(均P<0.05),14 d和28 d时D组与C组比较,皮层MAP-2阳性信号的表达升高(P<0.05)。结论:"头穴丛刺"能改善脑梗死大鼠的神经功能,增加缺血皮层MAP-2阳性信号的表达。 Objective To investigate the material base and underlying mechanism of the effect of cluster needling of scalp acupuncture on the neuronal plasticity in rats with focal cerebral infarction. Methods The model rats with acute cerebral infarction were made by blocking the middle cerebral artery with monofilament. One hundred and thirty two Wistar rats were randomly divided into 4 groups: sham operation group (A), model group (B), point-to- point scalp acupuncture group (C) and cluster-needling of scalp acupuncture group (D). Puncturing from "Baihui (GV 20)" to "Qubin (GB 7)" was used in group C. Cluster needling of scalp acupuncture was used in group D, in which needles were inserted forward and slantingly into "Baihui (GV 20)" and its left and right sides at 4 ram. In both groups, the treatment was carried out with rapid twirling reinforcing-reducing for 1 rain then retaining needle for 30 rain, once a day, 6 days in one course, for treating 4 courses. There was no treatment for group A and B. The change of neurological function was evaluated with P, ederson score, while the expression of microtubule-associated protein 2 (MAP-2) in the ische- mic penumbra was examined with immunohistochemistry (streptavidin-peroxidase method). Results In comparison,with group B, the score of neurological function in group D decreased on 7th day (P〈0. 05), while the scors in group C and D also decreased on 14th and 28th days (both P〈0.05). As compared with group C, the score of neurological function in group D obviously decreased on 28th day's (P〈O. 05). Comparing with group B, the expression of MAP-2 on the ischemic cortex was significantly increased in group D and C on 7th, 14th and 28th days (all P〈0. 05), however, this expression in group D was higher than that in group C on 14th and 28th days (P〈0.05). Conclusion Cluster needling of scalp acupunc- ture can improve the neurological function of rats with focal cerebral infarction, and increase the expression of MAP-2 in the Jscbemic penumbra,
