摘要
真菌SIPI-8917菌株的代谢产物具有抑制胆固醇生物合成酶活性,对其代谢产物进一步研究,应用丙酮浸泡、溶媒萃取、硅胶柱分离和LH20凝胶层析等方法,从其菌丝体中分离出另一种化合物,命名为SIPI-8917-Ⅴ。根据质谱(EI-MS、FAB和HRMS)数据,确定其分子式为:C28H44O1(分子量:396.3370),综合紫外光谱、红外光谱、1H-NMR、13C-NMR及HMBC和HMQC等图谱数据的解析,确定其结构为甾醇类化合物,旋光活性研究表明:与文献报道的麦角甾醇(ergosterol)结构一致,化学命名为:麦角-5,7,22E-三烯-3β-醇(ergosta-5,7,22E-trien-3β-ol)。
The metabolites of a positive strain named SIPI 8917 exhibited inhibitory activities on cholesterol biosynthetic enzyme. In the following studies, physiologically active compound SIPI 8917 Ⅴ was isolated from the mycelia of fungi SIPI 8917 by acetone soaking, solvent extraction, silica gel column, LH20 column chromatography etc .. Based on EI MS, FAB and HRMS, its formula was determined to be C 28 H 44 O (MW: 396.3370). By the analyses of UV, IR, 1H NMR, 13 C NMR, HMQC and HMBC etc . spectra data, the structure of SIPI 8917 Ⅴ was elucidated and was identical with that of ergosterol, and its chemical name is ergosta 5,7,22E trien 3β ol.
