摘要
为了解V79和V79-8两种细胞周期及调控因子的差异,用[3H]胸腺嘧啶核苷参入法检测两种细胞,发现V79细胞无可检测到的G2期;V79-8细胞则同时缺乏可检测到的G1和G2期。流式细胞光度术显示,V79G1期细胞明显多于V79-8,表明两者在G1期调控上存在差异。比较两者G1期周期调控因子的mRNA表达水平发现,V79-8cyclinD1,cyclinD3和Id表达水平明显高于V79。
V 79 and V 79 8 are two Chinese hamster lung fibroblast cell lines which proliferate very rapidly.It is verified through [ 3 H] thymidine incorporation assay that V 79 cells have non detectable G2 phase and V 79 8 cells have neither G1 nor G2 phase.With flow cytometry,we found that V 79 had obviously more G1 phase cells than V 79 8 ,which indicates that they are different in G1 phase regulation.Comparing the mRNA expression levels of several regulators in G1 phase,we also found that the expression levels of cyclin D1,D3 and Id in V 79 8 cells was obviously higher than that in V 79 cells.This might cause the G1 difference between V 79 and V 79 8 cells directly.
