固体豆粕为氮源根霉12~#产纤溶酶液体发酵条件的优化

摘要 以固体豆粕为氮源,通过单因素、Plackett-Burman试验设计和正交试验,从无机氮源、胰蛋白胨、初始pH值、接种量和发酵周期方面优化了中华根霉12#产纤溶酶的液体发酵条件。试验结果表明,该菌株液体发酵产纤溶酶的适宜培养基组成为:麸皮水5%,豆粕粉6%,NaNO30.2%,NH4Cl0.1%,胰蛋白胨2.5%,初始pH4.3;适宜的培养条件是:接种量20%,发酵周期60h,纤溶酶酶活力为182.00U/mL。 Liquid fermentation conditions for fibrinolysin production by Rhizopus chinesis 12# choosing soybean cake powder as organic nitrogen source was optimized by single-factor test, Plackett-Burman design and orthogonal test in terms of inorganic nitrogen source, tryptone, initial pH, inoculum and fermentation period. The results showed that the optimum medium consisted of wheat bran 5%, soybean cake power 6%, NaNO3 0.2%, NH4Cl 0.1% and tryptone 2.5% and adjusted pH value to 4.3. The optimum inoculum and fermentation duration were 20% and 60 h, respectively. Yield of fibrinolysin activity could reach 182.00 U/ml under optimum conditions.
