摘要
用大鼠杂交瘤技术,将人盲肠癌Hce-8693细胞免疫LOU/c大鼠,取其脾细胞与大鼠骨髓瘤IR983F细胞融合,共产生620株杂交瘤,从中筛选出分泌抗人大肠癌单抗杂交瘤195株,阳性率为31%。其中R19株分泌IgG2b型抗体,与人大肠癌Hce-8693和HT-29细胞呈强阳性反应,与人鼻咽癌CNE2细胞有弱阳性交叉反应,与正常人胚肺2BS细胞,人肝癌、人肺癌等共15种细胞无交叉反应,显示较强的特异性。用125I标记R19单抗在荷人盲肠癌裸鼠体内分布显示肿瘤部位特异性分布。
LOU/c rats were immunized with human cecum cancer Hce 8693 cells.The fusion of rat myeloma cells and immunized spleen cells produced 620 hybridomas.R19,one of the 195 positive hybridomas was selected for further study.Monoclonal antibody (McAb) R19,identified as IgG2b,showed strong reactivity with human cecum cancer Hce 8693 and colon cancer HT 29 cells,and week reactivity to human nasopharyngeal cancer CNE2 cells.However,no reactivity was found with human fetal lung 2BS,hepatoma,lung cancer,leukemia,stomach cancer,esophagus cancer,oral squamous cell carcinoma,breast cancer,murine colon cancer and murine melanoma.After injection of 125 I labeled R19 into nude mice bearing cecum cancer Hce 8693 and 131 I labeled R19 into nude mice bearing colon cancer HT 29,high accumulation was found in the tumor,and McAb R19 showed highly specific localization.
