摘要
采用纤维素酶、果胶酶和蛋白酶酶解法提取了海带褐藻胶,并对工艺进行了优化。试验结果表明:加入海带干重的2%纤维素酶,55℃(pH值4.5)水解20min,然后加入1%果胶酶60℃(pH值4.5)浸提1.5h,再加入1%蛋白酶,80℃(pH值8.0)水解3h,接着将浸提液加热至沸腾10min钝化酶活,最后加入1mol/L氯化钙(浸提液:氯化钙=4:1(v/v),离心沉淀后即得褐藻酸钙沉淀,加入次氯酸钠漂白后经盐酸酸化生成褐藻酸,干燥脱水后加入稀碳酸钠溶液即制得褐藻酸钠。
Alginates was extracted from laminaria by hydrolysis of cellulase, pectinase and protease and its extraction processes has been optimized. The results indicated that the optimal hydrolysis conditions were: 2% cellulase (at 55℃ and pH 4.5 for 20 min), 1% pectinase (at 60℃ and pH 4.5 for 1.5h) and 1% protease (at 80℃and pH 8.0 for 3h). And then all the enzymes were inactived by boiling solution for 10 min. 1 mol/L CaCl2 was finally added to the hydrolysis solution with the volume ratio of 1:4 (the hydrolysis solution: CaC12 solution). In the end, alginates was obtained by bleaching, acidification, drying and alkalization.
