转化人参二醇类皂苷为C-K的特异人参皂苷糖苷酶的纯化及性质 被引量:8

摘要 研究了真菌sp.g848p发酵产生的把人参二醇类皂苷PPD转化为人参皂苷C-K的特异的人参皂苷糖苷酶,及该酶的分离提纯。该酶经DEAE-Cellulose离子交换柱分离、聚丙烯酰胺凝胶电泳提纯,得到纯酶,其酶的分子质量约为74 ku。这种特异的人参皂苷糖苷酶能水解人参二醇类皂PPD生成稀有人参皂苷C-K,该酶最适反应时间为24 h,最适反应温度为45℃,最适pH为5.0。 The ginsenosidase transforming Protopanaxadiol ginsenosides into ginsenoside C-K was purified from the culture of Aspergillus sp. g848p strain. DEAE-Cellulose and SDS-polyacrylamide gel electrophoresis were used to purifying enzyme. The enzyme molecular mass was about 74 ku. The ginsenosidase can hydrolyze protopanaxadiol ginsenosides into ginsenoside C-K. The optimal condition of enzyme reaction is 24 h, 45 ℃, pH 5.0.
