黄芩苷对人外周血T细胞TCR Vβ mRNA表达的影响 被引量:2

摘要 目的研究黄芩苷对人外周血T细胞表面TCR VβmRNA表达的影响及其可能受体。方法采用尼龙毛柱法分离T淋巴细胞,将其分为空白对照组(T1)、黄芩苷组(T2)、抗体封闭用药组(T3)、抗体封闭对照组(T4),37℃培养48h,Trizol法提取总RNA,实时荧光RT-PCR法检测各组TCR VβmRNA表达。结果T2组较T1组TCR VβmRNA的表达明显增强(P(0.01);T3组较T2组TCR VβmRNA的表达明显减低(P(0.01);T3组较T4组TCR VβmRNA的表达增高(P>0.05)。结论黄芩苷能显著增加T细胞表面TCR VβmRNA的表达;T细胞受体(TCRαβ)是黄芩苷作用于T细胞的主要受体之一。 Objective To study the effect of baicalin on the mRNA expression of TCR vβ on the surface of human peripheral blood T cells and explore the possible receptor for baicalin. Methods T cells were separated by nylon fiber column method, and diuided into four groups: blank control group( T1 ) , baicalin group( T2 ) , antibody blocking 4 drug group( T3 ) and blocking control group(T4). After incubation for 48 h, the TCR vβ mRNA expression in different groups was detected by real -time fluorescent RT - PCR. Results 1. The TCR vβ expression of T2 group was significant higher than that of T1 group(P 〈 0.01 ). 2. The TCR vβ expression of T3 group was obviously decreased compared with T2 group(P 〈 0.01 ). 3. The TCR vβ expression of T3 group was higher than that of T4 group( P 〉 0.05 ). Conclusion Baicalin can improve the TCR vβ expression significantly and the TCR αβ is one of the major receptors for baicalin.
