补肾方对聋豚鼠耳蜗螺旋神经节细胞凋亡基因表达的影响 被引量:5

摘要 目的探讨金匮肾气丸对庆大霉素造成药物性耳聋豚鼠耳蜗螺旋神经节细胞凋亡调控基因表达的影响。方法豚鼠腹腔注射庆大霉素100 mg/(kg.d)制作耳聋动物模型,治疗组在注射庆大霉素后灌服金匮肾气丸水煎剂,实验结束后豚鼠断头处死,取耳蜗标本。观察指标:免疫组化Bax及Bc l-2的表达定量检测分析和电镜组织学观察。结果模型组Bax蛋白表达比正常组、治疗组表达显著增多(P<0.01)。模型组、治疗组Bc l-2蛋白表达均明显减少(P<0.01),治疗组Bc l-2蛋白表达高于模型组(P<0.01)。结论金匮肾气丸可通过抑制Bax蛋白表达,调节Bc l-2蛋白表达而达到对耳蜗螺旋神经节细胞凋亡的调控作用。 Objective To explore the protective effects of Jingui Shenqi Pill on apoptosis genes of spiral ganglion cells in guinea pigs with ototoxity by gentamycin; Methods The ototoxity model was induced by abdominal injection of 100 mg/( kg · d) gentamycin,while the treatment group took Jinguishenqi pill after injection of gentamyein, executed after the experiment, and the cochleas were gathered for detection. The observation indexes were as follows: the quantitative testing examination analysis and electro - microscopic observation of immunohistochemical Bax and Bcl - 2. Results Compared with other groups, model group had much higher expression of Bax(P 〈0. 01 ) ; the expression of Bcl -2 was obviously reduced both in treatment group and model group( P 〈 0.01 ), especially in model group( P 〈 0.01 ). Conclusion Jingui Shenqi Pill has effects on spiral ganglion ceils apoptosis by controling the expression of Bax and regulating the exnression of Bcl - 2.
