摘要
应用生物信息学的方法和工具对番茄LeNHX3蛋白质的理化性质、跨膜区域、疏水性/亲水性、二级结构、结构功能域、功能分类和同源性进行了分析,结果表明,番茄LeNHX3蛋白为疏水性稳定蛋白,包含一个高度保守的具有氨氯吡嗪咪结合位点的LFFIYLLPPI结构功能域,相对分子量为59.4kD,等电点为8.54,可能存在10个跨膜区域,蛋白质二级结构中的主要构成元件是α-螺旋和不规则卷曲;蛋白质同源性分析后,发现番茄LeNHX3蛋白和紫高杯花、矮牵牛花2种植物的Na+/H+反转运蛋白高度同源,序列相似性为90%。
The physical and chemical properties,transmembrane region,hydrophobicity/hydrophilicity,secondary structure,func- tional domain, functional category and homology tree of LeNHX3 protein were analyzed by using bioinformatics methods. Results showed LeNHX3 was hydrophobic protein containing an amiloride binding domain(LFFIYLLPPI),the calculated molecular mass was 59.4kD,theoretical isoelectric point was 8.54, there might be 10 transmembrane regions, the main components in the protein secondary structure were α-helix and random coil. Protein homology analysis showed LeNHX3 was close similar to Nierembergia hippomanica NhPurple and Petunia hybrida PhPurple by 90% identity respectively.
