脂多糖诱导小鼠腹腔巨噬细胞TNF-αmRNA、iNOSmRNA的表达 被引量:7

摘要 目的观察脂多糖对小鼠腹腔巨噬细胞肿瘤坏死因子-α以及诱导型一氧化氮合酶表达水平的影响。方法按常规方法获取小鼠腹腔巨噬细胞,随机分成两组:空白对照组及LPS组(分别含LPS 0.01,0.1,1.0,10mg/L)。采用荧光定量PCR方法测定肿瘤坏死因子-α及诱导型一氧化氮合酶的基因表达情况。结果经0.01μg/mL LPS刺激后细胞TNF-αmRNAi、NOS mRNA表达水平均明显升高,与空白对照组比较有显著性差异(P<0.01),并且在一定范围内呈剂量-效应关系。结论LPS可诱导小鼠巨噬细胞TNF-αmRNA i、NOS mRNA的表达。 Objective To observe the effects of lipopolysaceharide (LPS) induction on tumor necrosis faetor-a (TNF-α) and inducible nitric oxide synthase(iNOS)mRNA in mice peritoneal macrophages.Methods The ceils were divided into two groups randomly: control group (cells incubated without LPS), and LPS group (cells cultured in lnedium containing LPS with 0.01, 0.1, 1.0, 10mg/L respeetively),then cells were harvested after 24h incubation. The TNF-α and iNOS gene expression were detected by real-time reverse transcription-polymerase chain reaction (real time-PCR).Results The result analysis showed the expression of both iNOS mRNA and TNF-α mRNA in cells from 0.1mg/L LPS group was significantly increased compared with control group (P〈0.05). Conclusions LPS can significantly increase the expression of iNOS at gene level.
