金钗石斛总生物碱对四氧嘧啶所致糖尿病大鼠的保护作用 被引量:26

摘要 目的观察金钗石斛总生物碱(Dendrobium nobile Lindl.alkaloids,DNLA)对四氧嘧啶所致糖尿病大鼠的影响。方法体重200-220g的雄性SD大鼠60只随机分为空白、模型、总生物碱低、中、高剂量(40mg/kg、80mg/kg、160mg/kg)及二甲双胍(100mg/kg)组。预先灌胃给药1wk后腹腔注射四氧嘧啶(175mg/kg)制备高血糖大鼠模型,72h后取血测各组大鼠空腹血糖,并观察胰腺组织形态学变化。结果注射四氧嘧啶3d后,大鼠血糖较空白组明显增高(P<0.05),胰岛萎缩、数量减少,岛内细胞数减少;与模型组比较,DNLA(40mg/kg、80mg/kg、160mg/kg)及二甲双胍(100mg/kg)可明显降低大鼠血糖水平(P<0.05),胰岛数量较多,体积较大,岛内细胞数较多。结论DNLA可降低四氧嘧啶所致糖尿病大鼠血糖水平,其机制与DNLA对胰岛的保护作用有关。 Objective To investigate the effects of Dendrobium ncbile Lindl. alkaloids (DNLA) on alloxan-induced hyperglycemia in rats. Methods Spragye-Dawley rats are randomly divided into 6 groups as following: normal, model, DNLA groups (40mg/kg,80 mg/kg, 160mg/kg)and Mefformin group (75mg/kg). After DNLA administration by garage for one week, hyperglycemia was induced by intraperitoneal injection with alloxan at the dose of 175mg/kg,the blood glucose was determined by chemical method, and pancreas structure was observed by H.E. staining, respectively. Results The blood glucose became significantly increased, compared with control (P〈0.05), both DNLA and Mefformin could significantly decrease the blood glucose, compared with model (P〈0.05). Conclusions DNLA could significantly decrease the blood glucose in alloxan induced hyperglycemia rats, and its mechanism is due to protecting the pancreatic islet.
