摘要
目的:探讨肝癌组织中基质金属蛋白酶-2(MMP-2)和CD147的表达与临床病理特征的关系。方法:应用RT-PCR和免疫组化MaxvisonTM法,分别检测56例肝癌及癌旁组织中MMP-2 mRNA,MMP-2和CD147蛋白的表达情况。结果:MMP-2 mRNA及蛋白在肝癌和癌旁组织的表达差异有统计学意义(P<0.05),但肝癌组织MMP-2 mRNA和蛋白间差异无统计学意义(P>0.05);MMP-2和CD147蛋白在Ⅲ~Ⅳ期与Ⅰ~Ⅱ期、转移与无转移的肝癌组织中表达差异有统计学意义(P<0.05),肝癌组织中MMP-2蛋白和CD147蛋白表达的吻合度较强(κ=0.689,P=0.000)。结论:MMP-2和CD147的表达与肝癌的TNM分期密切相关,可作为判断肝癌侵袭性的指标。
Objective To explore the expressions of MMP-2 and CD147 in primary hepatocellular carcinoma(HCC) and their association with the clinicopathological features.Methods MMP-2 mRNA expression was detected by RT-PCR in the cancer tissues and the adjacent tissues from 56 patients,so were the protein expressions of MMP-2 and CD147 by immunohistochemitry.Results The expressions of MMP-2 mRNA and protein differed significantly between the cancer tissue and the adjacent tissue(P〈 0.05).In the cancer tissue,there was no significant difference between the expression of mRNA and that of protein in the cancer tissue(P〉 0.05);the protein expressions of MMP-2 and CD147 differed significantly between stages Ⅲ and Ⅳ disease and Ⅰ and Ⅱ disease,and between metastatic and nonmetastatic disease(P 〈0.05 for both comparisons);and the expression of MMP-2 protein agreed with that of CD147 protein(κ= 0.689,P = 0.000).Conclusions The expressions of MMP-2 and CD147 are closely correlated with TNM stage and can be used as predictors for determining tumor aggressive behavior in hepatocellular carcinoma.
