七叶皂苷A、B的制备液相色谱法分离和结构确证

Structure Identification and Purification of Sodium Aescinate by Preparative HPLC

目的建立制备型高效液相对β-七叶皂苷钠原料进行分离纯化制得七叶皂苷A(escin Ia)和七叶皂苷B(escin Ib)的方法。方法采用制备色谱柱waters HRC18;以乙腈-1%醋酸(28∶72)为流动相;流速为42mL.min-1,检测波长为220nm。结果用紫外、红外、质谱和核磁共振法确证结构,经分析型HPLC检查,纯度达98.0%以上。结论该法可靠、准确,适合于七叶皂苷A、B的制备、分离、纯化。

Objective To establish a preparative HPLC method for the purification of crude product.Methods The purification was performed on a Waters HRC18 column and mobile phase of acetonitrile-1%acetic acid（28∶72）.The flow rate was 40mL·min-1,and the determination wavelength was set at 220nm.Results The purity was over 98.0% by HPLC.The Structure identification by the data of UV、IR、1HNMR and 13CNMR.Conclusions The method is reliable,accurate and suitable for escin Ia,b preparation,separation and purification.
