摘要
目的观察p42丝裂原活化蛋白激酶(p42-MAPK)反义寡核苷酸对肾性高血压导致心肌肥厚大鼠p42-MAPK表达的影响。方法纳入健康清洁级SD大鼠30只,建立肾性高血压大鼠模型,分为假手术组、模型组、反义链组、错义链组及脂质体对照组。用脂质体将p42-MAPK反义寡核苷酸转染入相应组别大鼠,测定收缩压、左心室质量指数、心肌细胞直径和面积。免疫印迹法测定p42-MAPK表达。免疫沉淀法测定p42-MAPK活性。结果p42-MAPK反义寡核苷酸能显著降低模型组p42-MAPK表达及活性,下调肥厚重塑相关指标。结论p42-MAPK反义寡核苷酸可能是基因治疗心肌肥厚重塑的新靶点。
Objective To observe effects of p42-MAPK anti-ODN on renovascular hypertension-induced cardiomyocyte hypertrophy. Methods Into the health of clean grade SD rats were 30.Establish the model of renovascular hypertension. Animals were divided into sham-operation group, model group, anti-ODN group, mis-ODN group and lipofectamine group.With the liposome p42-MAPK antisense oligonucleotide was transfected into the appropriate group rats. As indexes of cardiac hypertrophy, SBP, left ventricular mass index, transdiameter of cardiomyocyte and cardiomyocyte area were measured. The expressions of p42-MAPK was assessed using Western blot; the activity of p42-MAPK was assessed by immuno-precipitation. Resulta Anti-ODN ofp42-MAPK decreased the indexes of cardiac hypertrophy, inhibited the expression and activity of p42-MAPK. Conlusion p42-MAPK anti-ODN could be a new target of gene therapy.
