甲苯磺酸二硫酸腺苷蛋氨酸的液相色谱分析

摘要 利用高效液相色谱测量甲苯磺酸二硫酸腺苷蛋氨酸含量,在高效毛细管电泳液相色谱一体机上考察了甲苯磺酸二硫酸腺苷蛋氨酸色谱行为,建立了甲苯磺酸二硫酸腺苷蛋氨酸的含量分析方法。实验表明:以甲酸铵—水—甲酸—甲醇(pH=3.0)+辛烷磺酸钠(对离子)混合溶液为流动相,流速为1.0 mL/min,C_(18)色谱柱为固定相,在检测波长为252 nm时,检测到甲苯磺酸二硫酸腺苷蛋氨酸浓度在0.005~0.25 mg/mL范围内与其色谱峰高成线性关系,最小检测限为0.001 mg/mL,平均相对标准偏差为0.338 1%,样品回收率在92.6%~101.1%之间。同时对甲苯磺酸二硫酸腺苷蛋氨酸的药品进行了定量分析,获得了满意结果。分析结果数据表明建立的甲苯磺酸二硫酸腺苷蛋氨酸的液相色谱分析方法是可靠的,具有一定实用价值。 In this work, quantitative analysis of S--adenocyl--L--methionine was proposed. The high performance liquid chromatographic behavior of toluenesulfonic acid disulfuric acid S--ade- nocyl--L--methionine (SAM) was researched in the high performance capillary electrophoresis liquid chromatography. This methods mainly uses C18 column by using ammonium formate-wate- methanol-sodium octane sulfonate ion pair mixture solution as mobile phase with 1.0 mL/min flow ratio and UV detection at 252 nm. The results showed that under the optimum conditions the peak intensity was linearly related to the concentration of SAM in the range from 0. 005 mg/mL to 0.25 mg/mI, with the detection limit of 0. 001 mg/mL. The samples recoveries ranged from 92.6% - 101.1% with the relative standard deviations less than 0. 3381% in the linear range. Quantitative analysis of S--adenocyl--L--methionine was studied and the good results were obtained.
