摘要
目的检测MMP-2在新生小鼠模型中的表达,探讨MMP-2在早产儿视网膜病变发病过程中的作用。方法将7日龄C57BL/6J小鼠置于75%±2%高氧环境中,5天后再置于正常空气环境中,建立高氧诱导的早产儿视网膜病变的小鼠模型。于鼠龄17天时处死动物并摘除眼球,应用免疫组化方法检测视网膜组织中MMP-2的表达情况。结果MMP-2在对照组和高氧组的积分光密度值分别为16.33±4.13和21.12±6.29,两者比较有显著性差异(t=3.160,P<0.01)。结论MMP-2可能对早产儿视网膜病变的新生血管生成有一定作用。
Objective This research using Oxygen Induced Retinopathy model examines the expressions of MMP-2 and inquires its contribution into retinopathy pathology of premature babies.Methods 7-day newborn C57BL/6J rats were raised in standardized incubator and exposed to(75±2)%oxygen concentration for 5 days,then returned to normal air,in order to establish a rat model of retinopathy pathology of premature babies.Rats were killed on the 17th day of life.Retinas were dissected and stained by immuno-histochemical method.Results IOD of MMP-2 was significantly increased 21.12±6.29 in the ROP group,and 16.33±4.13 in the control group.There was significant difference(t= 3.160,P0.01) in the level of MMP-2 between ROP group and control group. Conclusions It suggests that the existence of MMP-2 in a rat model do exert some effect on the generation of retinal neovascularization of premature babies' retinopathy pathology.
