毛脉山莴苣再生体系建立的研究 被引量:3

摘要 以生长非常旺盛的毛脉山莴苣生长点为外植体,以附加不同浓度的6-BA、IAA、IBA、NAA、2,4-D的1/2MS为基本培养基,进行了毛脉山莴苣芽生长、不定芽分化、试管苗生根、试管苗移栽与扦插和试管苗移植的研究。结果表明:芽生长的理想培养基为1/2MS+6-BA0.1mg/L+IAA0.2mg/L;不定芽分化培养的理想培养基MS+BA0.4mg/L+NAA0.1mg/L;试管苗生根培养和生根继代培养的理想培养基是1/2MS+NAA0.1mg/L+IAA0.4mg/L;河沙与炉灰渣是试管苗移栽和扦插的理想基质,移栽和扦插后25d的平均成活率分别为95%和91%;移植于山坡上的试管苗生长非常旺盛,根系发达,增加1倍左右;鲜茎叶收获量增加24%。 The growing points of Lacruca raddeana Maxim were used as explants to research on the point growth, the adventitious buds differentiation and the test-tube taking roots, transplanting, cutting with different concentration of 6-BA,IAA,IBA,NAA,2,4-D to the medium. The results indicated that 1/2 MS+6-BA 0.1mg/L+IAA 0.2 mg/L was the ideal medium for the growing of point, MS+BA 0.4 mg/ L+NAA 0.1 mg/L was ideal medium for differentiating adventitious buds. The optimum medium for taking roots and subculture was 1/2MS+NAA0.1 mg/L+IAA 0.4 mg/L. The survival ratio of test-tube plantlets was 95% and 91% respectively with the ideal groundmass-sands and cinder. The weight of roots had doubled and the weight of stems and leaves increased 24% .
