白藜芦醇对人胰腺癌细胞Capan-1增殖和细胞周期的影响 被引量:5

摘要 目的:探讨白藜芦醇(Resveratrol,Res)对人胰腺癌细胞Capan-1增殖和细胞周期的影响及变化规律。方法:通过不同浓度(0.128~500000nmol/L)Res作用于增殖期的Capan-1,检测5d各组细胞增殖率和细胞生长状态;通过增殖率曲线获得其半数生长抑制浓度(IC50)和杀伤浓度(killing concentration),并以此浓度再次作用细胞5d,检测并分析其细胞周期和细胞凋亡情况。结果:Res在16nmol/L时增殖率显著升高,而在15000nmol/L开始呈现抑制增长表现。Res对Capan-1的IC50为48.33μmol/L,细胞杀伤浓度为87.40μmol/L。流式细胞检测结果表明,Res浓度为48.33μmol/L时,G0/G1期细胞比例显著增高于空白对照组(P<0.01)。当Res浓度为87.40μmol/L时,G2/M期细胞比例显著低于空白对照组和48.33μmol/L组(P<0.01),且细胞凋亡率显著高于其他两组(P<0.01)。结论:Res对Capan-1的增殖影响存在浓度依赖性,分别表现为促进增殖、抑制增殖、促进凋亡3个阶段浓度,其中Res抑制细胞增殖和促进细胞凋亡的作用与其对细胞周期的阻断作用有关。
