曲古抑菌素A对HepG2细胞microRNA表达谱的影响 被引量:2

摘要 目的:探讨组蛋白去乙酰化酶(histone deacetylase,HDAC)抑制剂曲古抑菌素A(trichostatin A,TSA)对肝癌细胞HepG2microRNA(miRNA)表达谱的影响。方法:用浓度为0.2mol/L的TSA处理HepG2细胞24h后,采用miRNA表达谱芯片技术检测处理组和对照组细胞miRNA的表达,筛选出差异表达miRNA,并初步分析差异表达miRNA的可能生物学功能。结果:用浓度0.2mol/L的TSA处理HepG2细胞24h后,改变了HepG2细胞的miRNA表达。miRNA表达谱芯片结果显示,TSA处理后一共有8个miRNA表达发生了1.5倍以上的改变,其中有3个表达上调,5个表达下调,其中几个miRNA与肿瘤发生发展密切相关。结论:HDAC抑制剂TSA处理肝癌细胞HepG2后,影响了其miRNA的表达,这给TSA的抗癌机制研究提供了一些新的提示。但这些表达变化的miRNA是否在TSA抑制肝癌细胞增殖、诱导肝癌细胞分化的过程中具有重要的作用还有待更进一步的研究。
