氟烷和安氟醚对大鼠肝细胞显微与超微结构及组织化学的影响

摘要 选用Wester大白鼠28只,随机分为氟烷组及安氟醚组各12只、对照组4只。实验组用氟烷、安氟醚吸入麻醉,维持3小时。两组于麻醉后15天各取其中4只再作重复吸入麻醉(5天后处死),其余分别于10、20天处死,取肝脏作光镜、电镜、酶组化观察。结果发现,第10天氟烷组之肝损害较严重,20天逐步恢复;其中重复吸入组肝脏损害最严重;酶组化SDH、ATPase的变化与光镜及电镜的观察结果一致。在安氟醚组中,超微结构及酶组化仅有轻度变化,其重复吸入组变化办轻。本实验表明,氟烷对肝脏毒性作用比安氟醚明显,两药相比,后者较为安全,特别在短期内需重复用药者,选用安氟醚为佳。 In this paper 28 wester rats were chosen for the experiment of fluothane and enflurane hepatotoxicity. F group (n=12) was anaesthetized with fluothane by means of open inhalation method for 3 hours. 4 of them were re-exposed to fluothane on the 15th day after first aneasthesia and then sacrificed on the 5th day. The others were killed on the 10th and 20th days, respectively. E group (n=12) was anaesthetized with enflurane and all other experimental procedures were similar to the above. The results revealed severe damage of livers in the animals on the 10th day after they received fluothane, and the livers recovered gradually on the 20th day. In this group most serious damage of liver was observed in the animals of re-exposure to fluothane. Observations of enzyme histochemistry (SDH, ATPase) showed similarity to the above. Electron microscopic observation showed that the toxicity of fluothane on liver is stronger than that of enflurance. In comparision between the two drugs enflurane is a safe one; especially enflurance should be chosen for use when repeated inhalations are needed in a short time.
