地高辛素标记乙型肝炎病毒DNA探针的初步研究

摘要 将HBV/adw DNA克隆AM_6经SalⅠ内切酶酶切后作琼脂糖凝胶电泳回收,分别用随机引物法和缺口翻译法制备地高辛素、生物素标记HBVDNA探针。在DNA斑点杂交试验中,地高辛素、生物素标记HBVDNA探针分别检测到0.3pg和30pg的AM_6DNA。在100例临床拟诊病毒性肝炎患者血清标本中,地高辛素探针检出69例,生物素探针检出64例。初步研究表明,地高辛素标记HBVDNA是临床诊断的一个有力工具。 HBV/adw DNA clone AM_6 was linearized by Sal I digestion and further purified by gel electrophoresis. HBV DNA probes were labelled with digoxigenin and biotin by random priming and nick translation respectively.Digoxigenin-labelled and biotinylated HBV DNA probes detected 0.3 Pg and SO pg of AM_(?) DNA in DNA blot hybridization. A comparison study of 100sumples of sera from virus hepatitis patients using both probes showed that the digoxigenin probe picked up 69 positives while biotin probe picked up 64 positives. The preliminary study suggested that digoxigenin-labelled HBV DNA probe whould be a powerful tool for clinical diagnosis.
