氦-氖激光对人大颗粒淋巴细胞内嗜天青颗粒影响的图像分析研究 被引量:2

摘要 目的探讨氦-氖(HeNe)激光对人体的免疫刺激作用的机理。方法用3.1J/cm2、9.4J/cm2、25.0J/cm2和49.9J/cm2剂量的HeNe激光分别在体外辐照人外周血,细胞化学方法显示大颗粒淋巴细胞内嗜天青颗粒,并用图像分析系统对嗜天青颗粒作定量分析。结果3.1J/cm2辐照组的嗜天青颗粒数目、颗粒面积、颗粒面积与细胞面积比值和颗粒积分光密度四项参数值与对照组比较无明显变化;9.4J/cm2辐照组的四项参数值明显增高;25.0J/cm2辐照组的四项参数值进一步增高;49.9J/cm2辐照组的四项参数值较25.0J/cm2辐照组明显降低。结论适当剂量的HeNe激光能活化人大颗粒淋巴细胞内嗜天青颗粒,提高机体的免疫功能;最适激光剂量的选择是临床应用中应考虑的一个重要因素。 Objective To research He Ne laser stimulating immunity of human.Methods The human peripheral blood was irradiated in vitro by 3.1 J/cm 2, 9.4 J/cm 2, 25.0 J/cm 2 and 49.9 J/cm 2 of He Ne laser respectively.As the azurophilic granule of large granular lymphocytes was localized by a cytochemistry method, the image analysis system was adopted for quantitative analysis.Results The four indexes: number of azurophilic granule, area of granule, ratio of granule area to cell area and granule integral optical density at 3.1 J/cm 2 of He Ne laser irradiation group showed no difference in comparison with the control group.The indexes at 9.4 J/cm 2 group showed significant increase,the indexes at 25.0 J/cm 2 group showed more increase.But the indexes at 49.9 J/cm 2 group showed obvious decrease in comparison with those of 25.0 J/cm 2 group.Conclusion He Ne laser at appropriate dose may activate the azurophilic granule of large granular lymphocytes, raise human immunity.
