摘要
目的探讨BMSCs移植治疗肝硬化的最佳移植途径。方法1.取6周龄健康sD雌性大鼠股骨骨髓,以密度梯度离心和贴壁法分离纯化BMSCs。2.体重250—300g雌性SD大鼠60只,CCl4联合酒精制做肝硬化模型。3.取模型大鼠50只随机分为对照组,尾静脉移植组,肝脏移植组,脾脏移植组,门静脉移植组,每组10只。各实验组均移植DAPI标记的BMSCs。4.3周后比较各组大鼠肝功能。结果实验各组大鼠肝功能均明显改善,其中肝脏局部移植组效果优于其他各组,其他各组之间差异无统计学意义。结论通过肝脏局部移植BMSCs治疗肝硬化比经外周静脉、门静脉、脾脏更有优势。
Objective To investigate the therapeutic effect of BMSCs on liver cirrhosis induced by CC14 and to explore the best way to transplantation. Methods 1.The femur bone marrow was taken from healthy 6-week-old SD female rats,and the separation and purification of BMSCs were carried out by using density gradient centrifugation and adherence method. 2.Sixty healthy female SD rats weighted 250-300g were prepared for cirrhosis models induced by CC14 and alcohol. 3.The model rats were randomly divided into 5 groups: control group,tail vein transplant group,partial liver transplant group,spleen transplant group and portal vein transplant group, with 10 rats in each group. All experimental groups received DAPI labeled BMSCs transplant. 4. Three weeks after the transplantation, the liver function in rats was compared. Results The hver function of all the experimental groups was significantly improved compared with the control group,the partial liver transplant group being better than the other groups,with no significant difference between the other groups. Conclusion Partial liver BMSCs transplantation is better in liver cirrhosis than other transplantation channels.
